English: Inflammation mediators and many cellular and environmental stresses can induce the expression of JMJD3 via distinct signaling pathways. JMJD3 is recruited to chromatin through the interaction with certain transcription factors. JMJD3 enhances the transcription of
immune genes by demethylating the repressive H3K27me3 on their promoters and gene bodies. Moreover, JMJD3 can mediate an interaction between chromatin modifiers, and thus, it can enhance gene transcription in a demethylase-independent manner. JMJD3 can also control the nuclear localization of p53 and induce the expression of INK4 box genes by replacing the repressive polycomb complexes via a demethylation of H3K27me3. The induction of JMJD3 modifies the immune responses and triggers cellular senescence.
